I have a reparse rule from C:\1.txt to C:\2.txt.
When I open 1.txt it gets reparsed to 2.txt as it should.

Is there a way to determine on ReadFileC event whether this has been accessed from 1.txt to 2.txt?

Actually no.
Reparse rule working as follow: request open file - reject open with status reparse - open another file.
